A study of iga antibody response to different mycobacterium tuberculosis antigens in the diagnosis and monitoring of pulmonary tuberculosis BJID
Bezerra,Janaina Miranda; Beck,Sandra Trevisan; Kanunfre,Kelly Aparecida; Leite,Olavo Munhoz; Ferreira,Antônio Walter.
We evaluated the performance of the ELISA technique in the detection of IgA antibodies against different Mycobacterium tuberculosis antigenic preparations in serum samples from 49 patients with pulmonary tuberculosis collected before and after the start of specific treatment. The controls consisted of serum samples from healthy patients without any prior contact with the bacteria and serum samples from patients with other pneumopathies. Glycolipid antigen gave the best diagnostic performance, with a sensitivity of 88% and specificities varying from 88 to 100% in the control groups. These antigens constitute a powerful tool for the diagnosis and monitoring of patients with pulmonary tuberculosis.

Effect of Visfatin on the Structure and Immune Levels in the Small Intestine of LPS-Induced Rats International Journal of Morphology
Zhou,Ying; Cui,Lu; Yuan,Huairui; Xiao,Ke; Khan,Faheem Ahmed; Guo,Liang; Yang,Zhi; Song,Hui.
This study investigated the effects of visfatin on the structure and the immunity levels in the small intestine of LPS-induced rats. Forty Wistar male and female SPF rats were randomly and equally divided into four groups: the saline (control), vistfatin, lipopolysaccharide (LPS), and visfatin+LPS co-stimulated. The functions of visfatin in the intestinal mucosal immunity were investigated by examining the variation of tissue structure, inflammation and immunity-related proteins in the intestine of immunologically stressed rats using HE staining, ELISA, immunohistochemistry and Western Blot. The results showed that, when compared with the control group, the visfatin-treated group showed a decrease in the intestinal villus height and width, and a...

Genetic potential for an acute inflammatory response in IgA glomerulonephritis in mice BJMBR
Kurihara,R.S.; Yokoo,M.; Domingues,W.V.; Cabrera,W.H.; Ribeiro,O.G.; Ibañez,O.M.; Malheiros,D.A.; Barros,R.T.; de Almeida Prado,E.B..
Mice selected on the basis of an acute inflammatory response (AIR) can provide information about the immunopathological mechanisms of glomerulonephritis. We studied the differences between mice selected for a maximal AIR (AIRmax that attract more polymorphonuclear cells to the site of injury) or a minimal AIR (AIRmin that attract more mononuclear cells) in an experimental model of IgA nephropathy in order to investigate the effect of genetic background on glomerular disease progression and the participation of the monocyte chemoattractant protein-1 (MCP-1) chemokine. IgA nephropathy was induced by intraperitoneal ovalbumin injection and bile duct ligation in AIRmax and AIRmin mice. Histological changes, urinary protein/creatinine ratio, serum IgA levels,...

Immunological evaluation of the intestinal mucosa of broiler chicks treated with Lactobacillus Spp. and challenged with Salmonella Enteritidis Rev. Bras. Ciênc. Avic.
Okamoto,AS; Andreatti Filho,RL; Lima,ET; Pereira,REP; Menconi,A; Rocha,TS; Marietto-Gonçalves,GA.
This study aimed at the antibody production by intestinal mucosa of broilers chicks were orally inoculated with Lactobacillus spp. at one and/or 21 days of age, and subsequently challenged with Salmonella enterica, subspecies enterica, serotype Enteritidis (S. Enteritidis). A total number of 288 drug-free broiler chicks was divided into 6 groups (groups A, B, C, D, E, and F), according to age at Lactobacillus spp. inoculation and SE challenge. The intestinal mucosa immune response was determined as the production of immunoglobulin A against S. Enteritidis, and evaluated by the Enzyme-Linked Immunosorbent Assay (ELISA) technique in intestinal washing fluid. Groups treated with Lactobacillus spp. presented higher IgA production only when the chicks were...

Influence of probiotics on Candida presence and IgA anti-Candida in the oral cavity BJM
Santos,Agda Lima dos; Jorge,Antônio Olavo Cardoso; Santos,Silvana Soléo Ferreira dos; Silva,Célia Regina Gonçalves e; Leão,Mariella Vieira Pereira.
Probiotics are defined as microorganisms that promote benefits to host health, mainly by regulating resident microbiota. Disequilibrium in microbiota can favor the growth of opportunist microorganisms and the development of pathologies, like candidosis caused by yeasts of the Candida genus. This work evaluated whether probiotics consumption was able to influence a specific immunological response to Candida and the presence of these yeasts in the oral cavity. Saliva samples were collected from healthy individuals and plated in Dextrose Saboraud Agar with chloramphenicol. Individuals presenting Candida in the oral cavity used the probiotic Yakult LBâ for 20 days, after which new collections and identifications were performed. Anti-Candida IgA analysis was...

Lipid and acute-phase protein alterations in HIV-1 infected patients in the early stages of infection: correlation with CD4+ lymphocytes BJID
Treitinger,Arício; Spada,Celso; Silva,Laura Maria D. da; Hermes,Elizabeth Martins; Amaral,Jorge Antônio; Abdalla,Dulcineia Saes Parra.
Lipid and acute-phase protein alterations have been described in various infection diseases, and they have been recorded during the early stages of HIV infection. Lipid and acute-phase protein profiles also have been correlated with cellular immunological abnormalities. To document these correlations during HIV infection, we studied 75 HIV-infected patients and 26 HIV-negative controls. Patients were classified according to the criteria proposed by the Walter Reed Army Institute: as WR-1 (CD4 lymphocytes, 1154 ± 268/mm³), WR-2 (CD4, 793 ± 348/mm³) and WR3/4 (CD4, 287±75 mm³). Triglycerides, total cholesterol and HDL-cholesterol concentrations were measured by enzymatic methods. Immunoglobulins (IgA and IgG) and acute-phase proteins (haptoglobin, a1-acid...

Reduction of the amount of intestinal secretory IgA in fulminant hepatic failure BJMBR
Dong-yan,Liu; Weiguo,Jiang; Pei,Liu.
Intestinal barrier dysfunction plays an important role in spontaneous bacterial peritonitis. In the present study, changes in the intestinal barrier with regard to levels of secretory immunoglobulin A (SIgA) and its components were studied in fulminant hepatic failure (FHF). Immunohistochemistry and double immunofluorescent staining were used to detect intestinal IgA, the secretory component (SC) and SIgA in patients with FHF (20 patients) and in an animal model with FHF (120 mice). Real-time PCR was used to detect intestinal SC mRNA in the animal model with FHF. Intestinal SIgA, IgA, and SC staining in patients with FHF was significantly weaker than in the normal control group (30 patients). Intestinal IgA and SC staining was significantly weaker in the...

ZapA, a possible virulence factor from Proteus mirabilis exhibits broad protease substrate specificity BJMBR
Anéas,M.A.F.; Portaro,F.C.V.; Lebrun,I.; Juliano,L.; Palma,M.S.; Fernandes,B.L..
The opportunistic bacterium Proteus mirabilis secretes a metalloprotease, ZapA, considered to be one of its virulence factors due to its IgA-degrading activity. However, the substrate specificity of this enzyme has not yet been fully characterized. In the present study we used fluorescent peptides derived from bioactive peptides and the oxidized ß-chain of insulin to determine the enzyme specificity. The bradykinin- and dynorphin-derived peptides were cleaved at the single bonds Phe-Ser and Phe-Leu, with catalytic efficiencies of 291 and 13 mM/s, respectively. Besides confirming already published cleavage sites, a novel cleavage site was determined for the ß-chain of insulin (Val-Asn). Both the natural and the recombinant enzyme displayed the same broad...
